A Study on the Principle of Histoimmunoenzyme Staining



Abstract
The principle that antibodies can be used as histochemical reagents if suitable markers were linked to antibody molecules without impairing the capacity of antibody to react with specific antigens were recognized nearly 50 years ago (Marrack, 1934). Of the various dyes that were tried as markers, the first being fluorescein-labelled antibodies that were used to localized antigens (Coons et al. 1942). But the if technic had some disadvantages, so other labels were sought. Linking antibody to an enzyme, in lieu of fluorescein, proved most practical. Several enzymes were assessed in this system, in particular horseradish peroxidase, The PAP method was first utilized by Sternberger and colleagues at 1970. After then this method had been most widely used on the detection of antigens in tissue section. Because this technic has apparently high degree of sensitivity.
